Known as "400 in 4" in reference to the four days being taken to complete the journey, the ride will raise funds for the Westmead Children's Hospital burns unit.
For Bulli resident Troy Ingle, who works as a firefighter at Woollahra in Sydney, it will be the first time he has taken part in the event.
"I think the Great Dividing Range will be a challenge, there's a bit of uphill there," he said.
"I'm looking forward to the downhill ... unfortunately, it's not downhill the whole way.
"All of us, we've been putting in training for two or three months."
Mr Ingle encouraged people to donate at the website 400in4.org.
About 25 firefighters will take part in the ride, with about a quarter from the Illawarra.
Riders will stop in Gundagai, Yass and Goulburn, with the journey beginning on March 24.
The charity has raised more than $275,000 since its creation eight years ago.
