The closure is to allow Roads and Maritime Services to do a range of works including replacing a damaged drain.
Work will take place from 6pm Sunday, May 4, to midnight on Friday, May 9.
The work will mean the closure of a 400-metre stretch of Lawrence Hargrave Drive just north of Scarborough station to traffic in both directions.
Detours will be in place 24 hours with motorists travelling north and south of the worksite to use Lawrence Hargrave Drive, Princes Motorway, Mount Ousley Road and Bulli Pass.
Pedestrian access will be maintained throughout the week.
