A man and a woman,who were travelling in the same car, were killed in the crash.
The accident occurred on the Princes Highway at Broulee Road, near Waldrons Swamp Rest Area, police said.
There ages are not yet known.
The Princes Highway is closed in both directions in Moruya.
Northbound motorists are being diverted via North Head Road, George Bass Drive then Broulee Road.
Southbound motorists are being diverted via Broulee Road, George Bass Drive and North Head Road.
It follows an accident involving a car and motorbike in the vicinity last month.
It is the second Princes Highway crash in two days, with two vehicles lodged on the median strip near the Batemans Bay Industrial area on Wednesday morning.
