The 41 U bus route, to be run by Premier Illawarra, was one of two new routes announced in the June state budget.
The other, which runs between Flinders and Shell Cove, was also featured in the budget.
The Flinders bus, No 52, went into service in November 2014 and the Dapto bus will start running on January 27 - in time for the start of the new semester.
The new route will travel between Dapto station and the university and will feature 14 services each weekday.
"The new bus service has been made possible thanks to a $92 million investment by the NSW government for brand-new buses to replace older vehicles and provide new routes where they're needed across the state," Transport Minister Gladys Berejiklian said.
"With more and more houses going up in West Dapto every week, it's important we make sure there are improvements to public transport to keep up with demand.
"We want people to know they can leave their car at home and rely on trains and buses to get to where they need to go."
Timetable information for the new route can be found here.
