The 37-year-old woman died in hospital on Thursday night from head injuries received in the attack.
She was believed to be the estranged partner of the alleged offender.
The second woman, 53, suffered serious hand and arm injuries.
Police said the two women were attacked in a car on Risdon Road, New Town, about 3.15pm on Thursday.
A 73-year-old Huon Valley man was arrested at the scene.
He has been charged with murder and will appear in court on Friday.
The shooting happened just metres from a soccer oval, which was filled with junior players within an hour of the incident.
A gun was seized at the scene, and several shell casings were visible underneath the cars.
Police said the man was driving a black Mercedes four-wheel drive, while the two women were in a silver Peugeot hatchback.
Police allege the man fired several shots after stopping the Peugeot on Risdon Road.
A passing motorist restrained the man until police arrived at the scene.
Risdon Road was closed for several hours after the shooting.
Inspector Grant Twining said the incident seemed to only involve the man and the two women.
Inspector Twining urged any other witnesses to come forward.
"What we do request is that if there are people out there who have witnessed this, could they please go to Hobart Police Station and make themselves known so we can get their perspective," he said.
