Friends Kate Bateup and Nikki Groves contacted officers from the National Parks and Wildlife Service about 2pm on Wednesday, reporting they were lost.
They told officers they had walked on an unknown track east from Waterfall railway station, but contact with them was lost soon after.
The NSW Ambulance Service were assessing the women's health, but said neither would require transfer to hospital.
Officers from Police Rescue and the Sutherland Local Area Command had recommenced the search for the two women at 8am on Thursday.
The search was in addition to a nearby operation searching for 16-year-old Cassie Olczak, missing since Sunday after apparently leaving a train at Waterfall station. Police say there is no link between the two cases.
Ms Olczak's mother made an emotional appeal for any information about her daughter to be provided to police.
Police are urging anyone with information to call Crime Stoppers on 1800 333 000.
7.30am: A search for two women believed to be missing in the Royal National Park is due to resume at 8am, after the 21-year-olds reported themselves lost on Wednesday afternoon.
The search is in addition to a nearby operation searching for 16-year-old Cassie Olczak, missing since Sunday after apparently leaving a train at Waterfall station.
Officers from Police Rescue and the Sutherland Local Area Command will begin the search for the two women, Nikki Groves and Kate Bateup, who contacted officers from the National Parks and Wildlife Service about 2pm on Wednesday.
They told officers they had walked on an unknown track east from Waterfall Railway Station and had become lost.
Police have not been able to re-establish contact with the women since the call.
Sutherland police launched a search with the Lifesaver helicopter, the Rural Fire Service, the State Emergency Service and park rangers, which was suspended at 7pm due to darkness.
Police have not been able to confirm if the women are still lost, or were able to make their way out.
