The drivers were tested as part of an operation run by the Lake Illawarra Local Area Command held on January 24 and 25.
During the operation police performed 199 random drug tests, with the six positive readings for either cannabis or methamphetamine.
Officers at the Lake Illawarra command also conducted 868 random breath tests, with just one driver charged with drink-driving.
During the long weekend across the Southern Region, which includes both the Illawarra and South Coast and numerous inland area commands, 50 drivers blew a positive reading for alcohol out of 43,417 tests.
The total number of traffic infringements, excluding speeding, was 795 while police pulled over 706 drivers for breaking the speed limit. One person died in the region - at Barrengarry near Shoalhaven.
