Despite the absence of seven regular first-graders, the Mallee Bulls patiently chiselled out a 22-7 half-time lead then ran riot with six second-half tries.
Shamrocks showed they were up for the challenge in the first 20 minutes and led 7-0 after an intercept try to fullback Liam Vuik.
Bulls centre Tyler Aitkin hit back with a try while livewire five-eighth Leon Mason's penalty goal gave Uni a lead they never relinquished.
"We put a lot of pressure on them at set-piece time and we dominated them at scrum time, which limited them getting much ball to play with," University coach Shaun McCreedy said.
"I suppose we were a bit nervous leading up to the weekend. We had a lot of boys out and sometimes there can be a bit of a mental hangover from a tough game like last week against Avondale, but the boys rebounded well.
"That's the sign of a good team, especially to put on the number of points we did. It was a very pleasing effort.
"It just goes to show we've got a bit of depth. The boys that came up for us put their hands up and were counted when we needed them most."
Uni remained in second spot and host title-holders Camden this weekend.
"Both teams will have their [NSW] Country boys back so it should be a good game," McCreedy said.
''We want to hold onto that second spot for the rest of the year, so we have to keep winning. We'll manage this period with the injuries we've got and keep ploughing ahead.''
Kiama provided more hard evidence that they are serious about a long overdue return to the playoffs with a 22-19 road win over Camden.
The visitors built a 22-14 lead before the Rams scored an unconverted try in the dying stages.
"It's probably our most complete win of the year. We played a very smart game," Kiama coach Darryl Thomas said.
"They were down on troops but we had four key guys out as well. It was gutsy. We were talking before the game that we needed a boost.
"We were getting a bit flat but that's definitely the boost the boys need.
"They were pumped up afterwards, which they should be.
''We've got a few guys carrying injuries but we'll deal with that on Tuesday.''
Unbeaten leaders Avondale cut Shoalhaven to shreads, hitting the century mark in a 103-5 rout at Gerry Cappetta Oval.
The Wollongong Vikings moved back into the top four with a 62-22 road win over Bowral.
