Emergency services were called to the Hume Motorway at Pheasants Nest, south of Picton Road, about 8.15am on Wednesday following reports a car had veered off the road and rolled.
A number of police and ambulance resources, including a rescue helicopter, rushed to the scene.
The driver and sole occupant of a Jaguar, a 74-year-old Sydney man, was treated at the scene by paramedics before being taken to Liverpool Hospital, where he later died.
A witness told Fairfax Media it appeared the car rolled down a ditch near the southbound off-ramp from the motorway to the Pheasants Nest service centre.
Officers from the Camden Police District are investigating the crash.
A report will be prepared for the Coroner.
