The Tomerong accident occurred just after 3pm near Turpentine Road and resulted in a person being trapped in one of the vehicles.
The Princes Hwy is closed for motorists travelling southbound.
Live Traffic NSW is advising motorists to exercise caution, allow additional travel time and follow Police directions.
Further north at Bomaderry there are 5km queues for northbound traffic which is described as heavy.
Motorists are advised to allow an additional 25 minutes travel time.
There has also been a two car accident closing one of two southbound lanes on the Hume Hwy at Marulan near Highland Way.
Northbound traffic is also affected as the crash investigation unit, emergency services and Transport for NSW attend the scene.
At 3.15pm there was a 17km queue northbound and a 21km queue for southbound traffic.
Motorists are advised to avoid the area if they can and expect lengthy delays of up to two hours if they are travelling on the Hume Hwy.
