The South Sydney hooker produced two pieces of brilliance in the space of five minutes to blow the match open in the second half and hand his side an unassailable lead.
The first came with the Kiwis hot on the attack and throwing the ball around on the last tackle.
Eventually it was Cook who plucked the ball out of the air, before running 70m to cross for the first four-pointer of the second half.
Just five minutes later Cook, who was named man of the match, was in the thick of the action again, the hooker darting from dummy half before linking up with James Tedesco, who crossed untouched.
That play ended up being Cook's final involvement in the game, with the hooker handed an early rest and replaced by Dragons star Ben Hunt.
Cook's second-half brilliance came after an even start to the match, with both teams fighting to gain the upper hand in front of a vocal crowd of 18,104 at WIN Stadium.
While it was the Kiwis who held a slight territorial advantage throughout the early stages of the first half, it was Australia who opened the scoring, with Latrell Mitchell landing a penalty goal in the eighth minute.
The Kangaroos extended their lead 11 minutes later when Mitchell took an intercept before linking up with Josh Addo-Carr, who raced away to score under the posts.
Corrimal's Tyson Frizell was next to score, the St George Illawarra star beating two New Zealand defenders on the way to the try line.
Frizell's try came in a period of dominance for the Australians, with the forward pack rolling through the middle of the field.
Payne Haas made a significant impact off the bench, while Frizell's Dragons teammate Paul Vaughan was also busy in his Test debut.
Trailing 12-0, the Kiwis were desperate to hit back before the break and prevent the scoreline blowing out.
Eventually they received an opportunity to cross, and Charnze Nicoll-Klokstad made no mistake, the centre finishing off a Kiwis attacking shift.
With Shaun Johnson missing the conversion from out wide, the Australians entered the sheds leading 12-4.
With the match evenly poised at the break, both teams were determined to gain the upper hand in the opening phase of the second half.
Eventually it was Damien Cook who was able to break the game open and extend the margin out to 24-4 with 23 minutes to play.
The Kangaroos effectively closed out the match, with a late Daly Cherry-Evans penalty goal securing a 22-point victory.
