The Lighthouse Keepers fell to Dapto a week ago in a match where five of the top six batsmen reached double figures. Just one, however, scored a half-century.
With his side coming up against an in-form Uni, Maddinson said it's crucial someone produces a big score if they are to secure a victory.
"Uni's always a team we have tough battles with," Maddinson said. "We were good enough last year to beat them each time we played, this year they'll want to knock us off first time around.
"Everyone got a start last week and no one got out to good bowling, we got ourselves out. It's critical this week that if we get set, we go on with it."
University enter the match without their top two stars, captain Mitchell Hearn and fast bowler Rhys Voysey both playing at the Country Championships.
Maddinson, however, ensured his side will not be taking their opponents lightly.
"They're missing one of the best bats in the competition and probably the best bowler. They'll be looking for someone to step up in their place. We need to take full advantage of the fact they've got their two best players out."
Saturday's other match will see competition leaders Northern Districts take on defending premiers Balgownie and Corrimal host Keira.
Dapto will chase a third-straight win when they play Helensburgh, while Wests will face Port Kembla at Figtree Oval.
