The 12th seeds, the duo will play Lara Arruabarrena and Ons Jabeur in the third match on Court Seven.
Perez and Stosur are hoping to build on their performance at the Brisbane International, where they fell in the first round to Yang Zhaoxuan and Lyudmyla Kichenok 3-6, 7-6, 10-6.
Stosur enters Thursday's doubles clash having lost her first round singles match to Caty McNally, 6-1, 6-4 on Monday.
She has had far more success on the doubles court in Melbourne, however, and enters this year's event as a defending champion.
Perez missed out on a singles berth at the Australian Open, losing to Yanina Wickmayer in qualifying.
The emerging talent will also play mixed doubles, Perez to partner with fellow Australian Luke Saville.
