Think It Over was a leading contender for the race after winning the George Ryder Stakes.
However, a veterinary inspection led to the five-year-old gelding being taken out of the field, after a stewards meeting less than an hour before start time.
"Think It Over (scratched) at 3:45pm on veterinary advice after the gelding sustained multiple abrasions and laceration in the float en route to the course," Racing NSW stewards tweeted.
Parker has won two Group 1s, after Dark Dream won the 2018 Queensland Derby, only to be taken off him after the race.
His last Doncaster runner was in 2010, when Brilliant Light ran a game third behind Rangirangdoo.
