There will only be four reasons to leave your home during the lockdown and masks must be worn in both areas from 6pm Friday.
People have been encouraged to stay at home ahead of the lockdown taking effect.
Anybody who leaves the Perth and Peel area on Friday before midnight, or has left since April 17, will also need to wear a mask regardless of where they are in Western Australia.
The 54-year old man had quarantined for 14 days in Perth at the Mercure on the same floor where COVID-19 transmission cases have been reported.
A close contact of the man has also tested positive for COVID-19 after he stayed with their family in Kardinya on April 17. Other contacts of the family are being tested.
The man also stayed for three nights in a self-contained unit at St Catherine's College at the University of Western Australia and visited businesses in the Northbridge area. A more comprehensive list of potential exposure sites is expected to be provided by contact tracers later tonight.
The man subsequently travelled to Melbourne, with all passengers on the Qantas flight he travelled on told to self-isolate pending COVID-19 tests results.
Anzac Day services and sporting events have been cancelled and pubs and restaurants will only be allowed to do takeways.
