The Hearts booked their spot in the round five showdown with a 4-3 win over Forest Kilarney last month.
Rod Williams' side will enter the game full of confidence after gaining some revenge for last year's District League grand final on Saturday, securing a 3-0 win over Oak Flats at Unanderra Oval.
The Hearts are unbeaten so far in 2021 and sit in second spot on the league's ladder, only behind Shell Cove on goal difference.
Unanderra is one of three Football South Coast team still in this year's FFA Cup and victory this week would see them join Wollongong Olympic in the sixth round. Olympic booked their spot with a 2-1 win over Inner West Hawks last Tuesday.
Fellow Illawarra Premier League club Wollongong United will also look to advance when they travel to face Canterbury-Bankstown on Wednesday night.
