McKeon stopped the clock in 50.58 seconds, the same time as Sweden great Sarah Sjostrom recorded in 2017 over the four 25m laps.
She was 1.49 seconds faster than Sweden's Michelle Coleman, with Australian Madison Wilson third in 52.16.
It comes after McKeon won seven medals in Tokyo, four of them gold including the 100m and 50m freestyle events, to make her the most successful swimmer in Australian Games history, with 11 overall.
"It's a pretty good way to end the meeting," McKeon told FINA TV.
"To equal Sarah's record is a bit of an honour, really, because she's such an impressive racer and has definitely got a lot of records and world records.
"It's kind of nice to be close to that world record (50.25 seconds, held by Cate Campbell) as well."
