At 3.30am police and emergency services responded to reports of a single car rollover on the Tarlee to Kapunda Road.
A 53-year-old man from Kapunda, the sole occupant of the car, died at the scene.
The Tarlee to Kapunda Road was closed for several hours while Major Crash Investigators examined the circumstances of the crash, but it has since been reopened to traffic.
The man's death brings South Australia's road toll to 39, compared to 38 last year.
Police urge motorists to take care on the road's this long weekend and remind people to wear seatbelts and don't drink or drug drive.
