The third-placed Knights have won five straight matches, including a gutsy win over ladder leaders Shellharbour City Sharks, heading into the showdown at Kevin Walsh Oval.
Meanwhile, Jamberoo have slipped to fourth after falling to back-to-back defeats for the first time in 2017.
A win for either side would solidify their place near the top of the comp and keep the pressure on leading duo Shellharbour City and Gerringong.
City are one of three clubs to have the weekend off with a bye, alongside the Stingrays of Shellharbour and Albion Park-Oak Flats.
The Lions host Berry-Shoalhaven Heads at Michael Cronin Oval.
On Sunday, Milton-Ulladulla Bulldogs meet Nowra Bomaderry Jets while finals aspirants Warilla Lake South Gorillas face Port Kembla.
