Paramedics were called to the Sage Hotel shortly before 6am.
The two females were treated and transported to Wollongong Hospital, a NSW Ambulance spokesman confirmed.
Rooms at the Harbour Street hotel remained cordoned off later on Monday as forensic services officers examined the area.
A guest of the hotel told the Mercury he heard a female screaming some time after midnight and later noticed five police cars outside, and nearby rooms declared a crime scene.
Management at the hotel declined to comment on the incident, referring the Mercury’s inquiries to police.
Wollongong police have refused to confirm an investigation into a sexual assault is underway, citing protocol.
A spokeswoman for NSW Police said: "police are investigating an incident at a hotel in Wollongong and at this stage no further information is available”.
