Lehmann, the son of former Australian coach Darren Lehmann, will lead the Redbacks in the absence of regular skipper Travis Head, who will be on national duties.
"I want the boys to have fun, enjoy their cricket, execute their skills and feel comfortable playing the cricket we want to play," Lehmann told reporters on Wednesday.
The 26-year-old was chosen ahead of wicketkeeper-batsman Alex Carey to lead the Redbacks while Head pushes for a Test cap in Australia's series against Pakistan in the United Arab Emirates next month.
"We can expect that Jake, who displays good tactical and man management skills, will take to this new leadership role with energy and skill," SA's high performance manager Tim Nielsen said.
"It is a great opportunity for the leadership of the team to continue to broaden and Jake to take a significant step forward in his personal and cricketing development."
Lehmann, whose father was a long-serving SA captain, will start in the role when the Redbacks open their domestic one-day campaign against NSW on September 20 in Perth.
The Redbacks included fringe Test bowler Chadd Sayers, who was overlooked for the Pakistan series, in their 14-man squad for the initial two one-dayers.
