Figtree will travel to North Dalton Park to face reigning premiers Wollongong Lions in a grand final rematch.
While they will be the underdogs, coach Ryan said the Kangaroos had prepared well for the season opener.
"I think we've had six, seven weeks of training under our belt. Most of the boys have stayed pretty fit during the lockdown, they didn't have much else to do except go for runs which has been pretty handy getting back into it," Ryan said.
"I think we accept that the skills won't be quite as good as what we would normally have, but we're happy with the balance across our squad and happy to go up against them again as well. We have big aspirations for this season and that's the one team that has always stood in our way of ultimate success."
Ryan said he expected "typical Lions footy" from their opponents on Saturday.
"They're very well structured, very highly skilled with lots of experienced heads next to really good youth as well," he said.
"Ray [Burgess] is a really good coach, he's a good friend of mine as well, and I think three premierships in five years is usually testament to knowing what it takes to win a game of football."
Last year's wooden spooners Shellharbour Suns will host Northern District Tigers to kick off their AFL South Coast men's premier division campaign on Saturday.
Bomaderry takes on Kiama Power while Wollongong Bulldogs have the bye.
