Biden, who spent 36 years in the US Senate and eight years as vice president, is expected to recount his experience overcoming life's hardships including the deaths of his first wife and infant daughter in a car crash in the 1970s and the death of his son Beau from a brain tumour in 2015.
He is also expected to touch on his plans for dealing with the coronavirus pandemic, the resulting economic crisis and demands across the country for racial justice.
Biden, 77, officially became the Democratic presidential nominee on Tuesday.
Two former contenders for the Democratic nomination, Senator Cory Booker and former mayor Pete Buttigieg, will also speak during the final day of the convention.
The four-day event is taking place mostly online as a health precaution amid the coronavirus pandemic.
US President Donald Trump plans to campaign on Thursday in the key swing state of Pennsylvania, in a town close to Biden's birthplace.
Polling shows Biden ahead of Trump on the national level amid anger over the president's handling of the pandemic but the race will be most contentious in swing states.
The Republican Party convention will begin on August 24 and has also been pared back due to the coronavirus.
