Archedemus punched his ticket to the Provincial decider at Royal Randwick next month after saluting in the Newcastle heat on Saturday with another strong display.
The performance saw Archedemus firm in the TAB market from $4.50 to $4 favouritism to prevail on day two of The Championships.
The Lope De Vega five-year-old is undoubtedly now the horse to beat in the final, but Markwell hopes she can further strengthen her grip on the race.
Kembla's leading horsewoman now plans to target stablemate Mandylion at the Kembla Grange qualifier on March 23.
The Nicconi mare took out the same heat last year as a three-year-old.
"She will go to Kembla and she will be knocking on the door too," Markwell told Sky Thoroughbred Central.
Mandylion is currently a $15 TAB hope for the final and is a half sister to Archedemus.
Archedemus has quickly risen through the ranks in Markwell's stable, putting together five consecutive victories in dominant fashion.
Archedemus, ridden by Chad Lever, looked for a moment to be under pressure during her latest run in Newcastle but surged clear late to beat Damien Lane's Oakfield Missile and Oakfield Twilight.
"He is certainly tough," Markwell said.
"When he gets to the front he just looks like he is cantering. Like he is saying 'come up to me'.
"He always wins by a good margin so that is a good sign."
Meanwhile, the nominations for Friday's Kembla Grange Classic meeting will be released on Monday morning.
The Group 3 fillies feature is expected to attract another strong field.
