Heffernan outsprinted St George's Joanne Arancibia in the opening scratch to set the tone for the meeting.
She then outlasted Dole in the tempo race, before defying an Arancibia challenge in the final sprint of the elimination event.
It meant she had the maximum 120 points heading into the final leg, the points race, which Heffernan won to secure a perfect result.
"It was great to get out there and finally get a race in," Heffernan, who has been focusing on the road rather than track cycling, told Cycling NSW. "We had six girls, so it was a bit different racing than if it was a big field but everyone gave it their all. It was pretty tough out there.
"I'm doing more road this season, so I have a couple of country opens - there's Tolland (Wagga) next - and then I'm working towards the Tour of Brisbane and a few others."

With omnium victory already secured for Heffernan, Dole was left in a match race in the final points race with Arancibia, outsprinting her on the final lap to secure the Illawarra the top two places on the medal dais.
In other results, Sarah Cliff was fourth overall in the under 19 omnium with 118 points, behind Meg Marker and Ryan Brittan won bronze in the men's under 19 on 119, behind Dubbo's Dylan Eather.
