Picton Road is closed between Macarthur Road and Almond Street, around six kilometres from the Hume Highway interchange.
Two trucks crashed head-on about 5am on Friday, with one rolling and catching fire, police said.
One driver, who was thrown from his truck, died at the scene.
Initially police said they could not find the second driver.
However, his body was found around 7.30am inside the burnt-out cabin of his truck.
Crash Investigators are at the scene.
Police said the road is likely to remain closed for some hours.
Motorists on the Hume Motorway can use Appin Road.
B-Doubles can't use MacArthur Road and Wilton Road and will need to use Appin Road instead.
The section of Picton Road between the Hume Motorway and Princes Motorway is notorious for fatal accidents.
In February 2016 a man died and several other were injured not far from the scene of Friday's crash.
One of the worst crashes on the same stretch of road in recent years was in 2009, when five members of a family who had fled Afghanistan in search of a new life were killed in a crash.
